They must do it at the queueing level, I totally agree with you that doing it at the character select screen would just lead to throwing.
As a support main who dabbles with tanks there is nothing I want more in the game than a 2-2-2 lock. It would bring a lot more structure to the game and be so much less frustrating, at least for the current reasons. I'm sure the new system will have it's own problems but I'm much happier to deal with those than being solo healer in way too many of my games.
